Valaciclovir: Side Effects Valaciclovir is the L-valyl ester of aciclovir. After oral administration, it is rapidly and extensively converted to aciclovir by first-pass metabolism, resulting in plasma aciclovir concentrations previously only attainable with intravenous administration.
